Paxillin is a cytoskeletal protein involved in actin-membrane attachment at sites of cell adhesion to the extracellular matrix (focal adhesion) . Extensive tyrosine phosphorylation occurs during integrin-mediated cell adhesion.mbryonic development.ibroblast transformation and following stimulation of cells by mitogens that operate through the 7TM family of G-protein-coupled receptors . Paxillin binds in vitro to the focal adhesion protein vinculin.s well as to the SH3 domain of c-Src.nd.hen tyrosine phosphorylated.o the SH2 domain of v-Crk . An N-terminal region has been identified that supports the binding of both vinculin and the focal adhesion tyrosine kinase.p125Fak .Paxillin is a 68 kDa protein containing multiple domains.ncluding four tandem C-terminal LIM domains (each of which binds 2 zinc ions); an N-terminal proline-rich domain.hich contains a consensus SH3 binding site; and three potential Crk-SH2 binding sites . The predicted structure of paxillin suggests that it is a unique cytoskeletal protein capable of interaction with a variety of intracellular signalling and structural molecules important in growth control and the regulation of cytoskeletal organisation .

Zyxin and paxillin are exceptions in that they contains respectively three and four LIM domains at their C-terminal extremity. In apterous.sl-1.H-2.in-11.im-1 to lim-3.mx-1 and ceh-14 and mec-3 there is a homeobox domain some 50 to 95 amino acids after the LIM domains.In the LIM domain.here are seven conserved cysteine residues and a histidine. The arrangement followed by these conserved residues is C-x(2)-C-x(16.3)-H-x(2)-[CH]-x(2)-C-x(2)-C-x(16.1)-C-x(2.)-[CHD]. The LIM domain binds two zinc ions . LIM does not bind DNA.ather it seems to act as an interface for protein-protein interaction.
