Higher eukaryotes have many distinct esterases. Among the different types arethose which act on carboxylic esters (). Carboxyl-esterases havebeen classified into three categories (A. and C) on the basis ofdifferential patterns of inhibition by organophosphates. The sequence of anumber of type-B carboxylesterases indicates that the majority are evolutionary related. As is the case for lipases and serine proteases.he catalytic apparatus ofesterases involves three residues (catalytic triad): a serine. glutamate oraspartate and a histidine.

Neuroligins constitute a family of brain neurone-specific membrane proteinswhose structural and biochemical characteristics are indicative of a role inheterotypic cell adhesion . Functionally.hey bind to the extracellulardomains of a subset of beta-neurexins in a Ca2+-dependent manner. They havebeen localised to the postsynaptic membranes of excitatory synapses .This.ogether with the fact that their C-termini likely bind to PSD-95 (aPDZ-domain protein enriched in postsynaptic densities).uggests that theymay be involved in the recruitment of proteins (such as receptors.onchannels and signal-transduction molecules) to synaptic sites of celladhesion.To date.hree neuroligins have been reported.esignated neuroligin 1-3 .Multiple sequence alignment has revealed them to be highly similar.haringoverall ~52% identity. The similarity is distributed over the whole proteinsequence.ith the extracellular and transmembrane (TM) domains being moreconserved than the cytoplasmic regions. They are composed of five domains:an N-terminal cleaved signal sequence; a large extracellular region homologous to esterases; a linker between the TM domain and the esterase homology domain that may be O-glycosylated; a single TM domain; and acytoplasmic C-terminal tail. Further sequence comparisons place theneuroligins into the large family of esterase homology domain proteins.hich includes thyroglobulin.cetylcholinesterase.nd gliotactin.lthough the neuroligins are themselves catalytically inactive.
