Domain Architecture (Details)
InterPro domains unassigned to SynO:
Elongation factors belong to a family of proteins that promote the GTP-dependent binding of aminoacyltRNA to the A site of ribosomes during protein biosynthesis.nd catalyse the translocation of thesynthesised protein chain from the A to the P site. The proteins are all relatively similar in the vicinity oftheir C-termini.nd are also highly similar to a range of proteins that includes the nodulation Q protein fromRhizobium meliloti.acterial tetracycline resistance proteins and the omnipotent suppressorprotein 2 from yeast. In both prokaryotes and eukaryotes.here are three distinct types of elongationfactors.F-1alpha (EF-Tu).hich binds GTP and an aminoacyl-tRNAand delivers the latter to the A site ofribosomes; EF-1beta (EF-Ts).hich interacts with EF-1a/EF-Tu to displace GDP and thus allows theregeneration of GTP-EF-1a; and EF-2 (EF-G).hich binds GTP and peptidyl-tRNA and translocates thelatter from the A site to the P site. In EF-1-alpha. specific region has been shown to beinvolved in a conformational change mediated by the hydrolysis of GTP to GDP. This region is conservedin both EF-1alpha/EF-Tu as well as EF-2/EF-G and thus seems typical for GTP-dependent proteins whichbind non-initiator tRNAs to the ribosome. The GTP-binding protein synthesis factor family also includes theeukaryotic peptide chain release factor GTP-binding subunits and prokaryotic peptide chainrelease factor 3 (RF-3) ; the prokaryotic GTP-binding protein lepA and its homolog in yeast(GUF1) and Caenorhabditis elegans (ZK1236.1); yeast HBS1 ; rat statin S1 ; and the prokaryoticselenocysteine-specific elongation factor selB .
  IPR000795:Protein synthesis factor, GTP-binding
Elongation factor Tu consists of three structural domains.his is the second domain. This second domain adopts a beta barrel structure.nd is involved in binding to charged tRNA . This domain is also found in other proteins such as elongation factor G and translation initiation factor IF-2. This domain is structurally related to .nd in fact has weak sequence matches to this domain.
  IPR004161:Elongation factor Tu, domain 2
Initiation factor 2 (IF-2) (gene infB) is one of the three factorsrequired for the initiation of protein biosynthesis in bacteria. IF-2promotes the GTP-dependent binding of the initiator tRNA to the small subunitof the ribosome. IF-2 is a protein of about 70 to 95 Kd which contains acentral GTP-binding domain flanked by a highly variable N-terminal domain anda more conserved C-terminal domain.Bacterial IF-2 is structurally and functionally related to eukaryoticmitochondrial IF-2 (IF-2(mt)) as well as to algal and plants chloroplastIF-2 (IF-2(chl)). Both IF-2(mt) and IF-2(chl) are encoded by nuclear genes andare produced as precursor proteins with a transit peptide. An exception arered algae where IF-2(chl) is encoded by the plastid genome .
  IPR000178:Initiation factor 2
Proteins with a small GTP-binding domain include Ras.hoA.ab11.ranslation elongation factor G.ranslation initiation factor IF-2.etratcycline resistance protein TetM.DC42.ra.DP-ribosylation factors.dhF.nd many others . In some proteins the domain occurs more than once. Among them there is a large number of small GTP-binding proteins and related domains in larger proteins. Note that the alpha chains of heterotrimeric G proteins are larger proteins in which the NKXD motif is separated from the GxxxxGK[ST] motif (P-loop) by a long insert and are not easily detected by this model.
  IPR005225:Small GTP-binding protein domain
A beta barrel of circularly permuted topology is found in many transcription proteins.ncluding initiation and elongation factors.nd also some ribosomal proteins.lthough in these cases the fold is elaborated with additional structures. The beta barrel domain is represented by domain 2 of the elongation factors EF-Tu and eEF1A .oth of which function to recognize and transport aminoacyl-tRNA to the acceptor (A) site of the ribosome during the elongation process.nd of EF-G .hich functions in translocating the peptidyl tRNA from the A site to the peptidyl (P) site. This domain is also present in initiation factors.n domain 2 of eIF2 gamma subunit .nd domains 2 and 4 of IF2/eIF5B .oth of which function to transport the initiator methionyl-tRNA to the ribosome. This beta barrel domain may be involved in interactions with the switch 2 region to stabilise the relative orientations of the domains.hich undergo functionally important conformational changes between GTP- and GDP-bound states. The SSF signature in this entry is currently under review. Please be aware that some of the protein hits may be false positives.
