Elongation factors belong to a family of proteins that promote the GTP-dependent binding of aminoacyltRNA to the A site of ribosomes during protein biosynthesis.nd catalyse the translocation of thesynthesised protein chain from the A to the P site. The proteins are all relatively similar in the vicinity oftheir C-termini.nd are also highly similar to a range of proteins that includes the nodulation Q protein fromRhizobium meliloti.acterial tetracycline resistance proteins and the omnipotent suppressorprotein 2 from yeast. In both prokaryotes and are three distinct types of elongationfactors.F-1alpha (EF-Tu).hich binds GTP and an aminoacyl-tRNAand delivers the latter to the A site ofribosomes; EF-1beta (EF-Ts).hich interacts with EF-1a/EF-Tu to displace GDP and thus allows theregeneration of GTP-EF-1a; and EF-2 (EF-G).hich binds GTP and peptidyl-tRNA and translocates thelatter from the A site to the P site. In EF-1-alpha. specific region has been shown to beinvolved in a conformational change mediated by the hydrolysis of GTP to GDP. This region is conservedin both EF-1alpha/EF-Tu as well as EF-2/EF-G and thus seems typical for GTP-dependent proteins whichbind non-initiator tRNAs to the ribosome. The GTP-binding protein synthesis factor family also includes theeukaryotic peptide chain release factor GTP-binding subunits and prokaryotic peptide chainrelease factor 3 (RF-3) ; the prokaryotic GTP-binding protein lepA and its homolog in yeast(GUF1) and Caenorhabditis elegans (ZK1236.1); yeast HBS1 ; rat statin S1 ; and the prokaryoticselenocysteine-specific elongation factor selB .

Elongation factor Tu consists of three structural domains.his is the second domain. This second domain adopts a beta barrel structure.nd is involved in binding to charged tRNA . This domain is also found in other proteins such as elongation factor G and translation initiation factor IF-2. This domain is structurally related to .nd in fact has weak sequence matches to this domain.

Initiation factor 2 (IF-2) (gene infB) is one of the three factorsrequired for the initiation of protein biosynthesis in bacteria. IF-2promotes the GTP-dependent binding of the initiator tRNA to the small subunitof the ribosome. IF-2 is a protein of about 70 to 95 Kd which contains acentral GTP-binding domain flanked by a highly variable N-terminal domain anda more conserved C-terminal domain.Bacterial IF-2 is structurally and functionally related to eukaryoticmitochondrial IF-2 (IF-2(mt)) as well as to algal and plants chloroplastIF-2 (IF-2(chl)).
