Many calcium-binding proteins belong to the same evolutionary family and share a type of calcium-binding domain known as the EF-hand. This type of domain consists of a twelve residue loop flanked on both side by a twelve residue alpha-helical domain. In an EF-hand loop the calcium ion is coordinated in a pentagonal bipyramidal configuration. The six residues involved in the binding are in positions 1.... and 12; these residues are denoted by X...Y.X and -Z. The invariant Glu or Asp at position 12 provides two oxygens for liganding Ca (bidentate ligand).

The protozoan parasite that causes Chagas disease.rypanosoma cruzi.ontains a 24kDa protein that is recognised by antiera from both humans andexperimental animals infected with this organism. Near its C-terminus are two regions that have sequence similarity with E-F hand Ca2+-binding proteins . Indeed.he native trypanosome protein exhibits low Ca2+-binding capacity and high Ca2+-binding affinity.onsistent with binding via E-F hand structures. Immunofluorescence assays have suggested that the protein is localised to the trypanosomes flagellum. This observation.oupled with the proteins Ca2+-binding properties.uggests that it may participate in molecular processes associated with the high motility of the parasite . A set of similar 24 kDa proteins.ermed calflagins.re contained within the flagellum of Trypanosoma brucei. These contain three EF-hand Ca2+- binding domains and one degenerate EF-hand motif .

The fold consists of four helices and an open array of two hairpins.
